介紹

以急性發(fā)熱性呼吸道癥狀為特征的退伍軍人癥是由吸入含有嗜肺軍團(tuán)菌的霧化水引起的。嗜肺軍團(tuán)菌血清組1zui常從患者中分離（1,2）。
IMMUNOCATCHTM軍團(tuán)菌（Legionella）是一種側(cè)流免疫層析法，可通過(guò)快速可靠地檢測(cè)尿樣中的抗原，對(duì)嗜肺軍團(tuán)菌血清型1感染進(jìn)行早期診斷。

原理

本產(chǎn)品是一種試劑盒，設(shè)計(jì)用于免疫層析法檢測(cè)尿樣中嗜肺軍團(tuán)菌血清型1 LPS抗原。
當(dāng)樣品通過(guò)毛細(xì)作用遷移通過(guò)結(jié)合墊時(shí)，樣品中的軍團(tuán)菌抗原與綴合至膠體顆粒的抗軍團(tuán)菌抗體結(jié)合形成抗原 - 抗體復(fù)合物。抗原 - 抗體復(fù)合物在試紙條上遷移，與固定在膜上的抗軍團(tuán)菌抗體特異性結(jié)合而產(chǎn)生紅線。然后通過(guò)肉眼觀察紅線的存在來(lái)確定軍團(tuán)菌抗原的存在。
此外，抗-Legionella抗體偶聯(lián)的膠體金在反應(yīng)中不被消耗，與固定在對(duì)照線上的抗免疫球蛋白抗體結(jié)合產(chǎn)生紅線，表明在測(cè)試條上的反應(yīng)是成功的。

【檢測(cè)方法】

1）從鋁袋中取出所需數(shù)量的磁帶，并將它們放在水平面上。

2）使用定量移液器慢慢地吸取90μL或更多的樣品。 通過(guò)一次移液操作，將全部樣品滴入盒子的樣品位置。 滴下后，剩余的標(biāo)本留在吸管內(nèi)。

3）在室溫（15-30°C）下將盒子放置15分鐘，

4）檢查盒子評(píng)估地點(diǎn)是否有線（標(biāo)有“C”和“T”）。

結(jié)果

應(yīng)該在反應(yīng)開(kāi)始15分鐘后及時(shí)進(jìn)行評(píng)估。 檢查評(píng)估地點(diǎn)，并通過(guò)下面描述的紅線來(lái)評(píng)估結(jié)果。

軍團(tuán)菌尿抗原檢測(cè)試劑盒

繁殖力強(qiáng)，繁殖水溫以24℃為宜，繁殖周期約7天左右，每年可繁殖6-8次。雌雄魚(yú)的交配行為受光刺激，可以通過(guò)調(diào)控光周期或控制雌雄魚(yú)的接觸而控制產(chǎn)卵時(shí)間。每尾雌魚(yú)每次產(chǎn)卵300粒左右，體型較大者有時(shí)可以產(chǎn)上千粒。卵子體外受精，體外發(fā)育，胚胎發(fā)育同步且速度快，胚體透明。受精卵在28.5℃培養(yǎng)條件下約40min完成*次有絲分裂，之后大約每間隔15 min分裂一次；24 h后，主要組織器官原基形成，各個(gè)腦室、眼睛、耳、血細(xì)胞、體節(jié)等均清楚可見(jiàn)。大約兩天后仔魚(yú)卵黃囊消失，可游動(dòng)攝食。發(fā)育溫度要求在25-31℃之間。幼魚(yú)約2個(gè)月后可辨雌雄，5個(gè)月可達(dá)性成熟。
二、易于飼養(yǎng)管理
斑馬魚(yú)飼養(yǎng)條件比較粗放，其細(xì)菌溫和，活潑好動(dòng)，喜在上層水域活動(dòng)覓食，對(duì)餌料不挑剔，各種魚(yú)蟲(chóng)及人工飼料均可投喂。
目前，國(guó)內(nèi)外已經(jīng)發(fā)展了多種斑馬魚(yú)養(yǎng)殖設(shè)備。國(guó)內(nèi)已建成自動(dòng)化控溫控光和水循環(huán)處理的斑馬魚(yú)養(yǎng)殖系統(tǒng)，并建立了較為完善的相關(guān)研究技術(shù)平臺(tái)。建立了國(guó)家斑馬魚(yú)模式動(dòng)物中心(http://www.zfish。。ｃｎ/)，為國(guó)內(nèi)研究人員提供斑馬魚(yú)資源、技術(shù)和信息服務(wù) 。
三、分子遺傳學(xué)特征
斑馬魚(yú)有25對(duì)染色體連鎖群，已完成組裝的序列總長(zhǎng)度為1.63×109 bp，已注釋的編碼基因21503個(gè)、RNA基因3421個(gè)。
斑馬魚(yú)的基因組中含有約30000個(gè)基因，這個(gè)數(shù)目與人的差不多，而且它的許多基因與人體存在對(duì)應(yīng)關(guān)系，更為難得的是，斑馬魚(yú)的腫瘤情況與人極為類似，因此，斑馬魚(yú)作為一種理想的分子生物學(xué)模式生物，擁有其它脊椎動(dòng)物所*的優(yōu)點(diǎn)。
斑馬魚(yú)的細(xì)胞標(biāo)記技術(shù)、組織移植技術(shù)、單倍體育種技術(shù)、隨機(jī)及靶基因定向誘變技術(shù)、轉(zhuǎn)基因技術(shù)、基因過(guò)量表達(dá)技術(shù)、基因活性抑制技術(shù)、體細(xì)胞克隆技術(shù)等已經(jīng)成熟。如：利用斑馬魚(yú)可以很方便地進(jìn)行在低等模式生物(線蟲(chóng)、果蠅)上進(jìn)行的細(xì)胞標(biāo)記和細(xì)胞譜系跟蹤，也可像利用爪蟾一樣做胚胎的細(xì)胞移植。此外，還可培育單倍體及進(jìn)行基因組倍增。
