以急性發(fā)熱性呼吸道癥狀為特征的退伍軍人癥是由吸入含有嗜肺軍團(tuán)菌的霧化水引起的。嗜肺軍團(tuán)菌血清組1zui常從患者中分離（1,2）。
IMMUNOCATCHTM軍團(tuán)菌（Legionella）是一種側(cè)流免疫層析法，可通過快速可靠地檢測尿樣中的抗原，對嗜肺軍團(tuán)菌血清型1感染進(jìn)行早期診斷。

原理

本產(chǎn)品是一種試劑盒，設(shè)計(jì)用于免疫層析法檢測尿樣中嗜肺軍團(tuán)菌血清型1 LPS抗原。
當(dāng)樣品通過毛細(xì)作用遷移通過結(jié)合墊時(shí)，樣品中的軍團(tuán)菌抗原與綴合至膠體顆粒的抗軍團(tuán)菌抗體結(jié)合形成抗原 - 抗體復(fù)合物?？乖?- 抗體復(fù)合物在試紙條上遷移，與固定在膜上的抗軍團(tuán)菌抗體特異性結(jié)合而產(chǎn)生紅線。然后通過肉眼觀察紅線的存在來確定軍團(tuán)菌抗原的存在。
此外，抗-Legionella抗體偶聯(lián)的膠體金在反應(yīng)中不被消耗，與固定在對照線上的抗免疫球蛋白抗體結(jié)合產(chǎn)生紅線，表明在測試條上的反應(yīng)是成功的。

【檢測方法】

1）從鋁袋中取出所需數(shù)量的磁帶，并將它們放在水平面上。

2）使用定量移液器慢慢地吸取90μL或更多的樣品。 通過一次移液操作，將全部樣品滴入盒子的樣品位置。 滴下后，剩余的標(biāo)本留在吸管內(nèi)。

3）在室溫（15-30°C）下將盒子放置15分鐘，

4）檢查盒子評估地點(diǎn)是否有線（標(biāo)有“C”和“T”）。

結(jié)果

應(yīng)該在反應(yīng)開始15分鐘后及時(shí)進(jìn)行評估。 檢查評估地點(diǎn)，并通過下面描述的紅線來評估結(jié)果。
1）正面：紅色控制線和測試線均在評估現(xiàn)場。 當(dāng)兩條線在反應(yīng)時(shí)間結(jié)束之前出現(xiàn)時(shí)結(jié)果是肯定的。
2）否定的：評估地點(diǎn)只有一條紅色的控制線。

1、單向瓊脂擴(kuò)散試驗(yàn)：是一種常用的定量檢測抗原的方法。將適量抗體與瓊脂混勻，澆注成板，凝固后，在板上打孔，孔中加入抗原，抗原就會向孔的四周擴(kuò)散，邊擴(kuò)散邊與瓊脂中的抗體結(jié)合。一定時(shí)間后，在兩者比例適當(dāng)處形成白色沉淀環(huán)。沉淀環(huán)的直徑與抗原的濃度成正比。如事先用不同濃度的標(biāo)準(zhǔn)抗原制成標(biāo)準(zhǔn)曲線，則從曲線中可求出標(biāo)本中抗原的含量。單向瓊脂擴(kuò)散試驗(yàn)主要用于檢測標(biāo)本中各種免疫球蛋白和血清中各種補(bǔ)體成分的含量，敏感性很高。
2、雙向瓊脂擴(kuò)散試驗(yàn)：是將半固體瓊脂傾注于平皿內(nèi)或玻片上，待其凝固后，在瓊脂板上打孔，將抗原、抗體分別注入小孔內(nèi)，使兩者相互擴(kuò)散。如果抗原、抗體相互對應(yīng)，濃度、比例適當(dāng)，則一定時(shí)間后，在抗原、抗體孔之間出現(xiàn)清晰可見的沉淀線。雙向瓊脂擴(kuò)散法可用來分析溶液中的多種抗原。一對抗原、抗體系統(tǒng)只能形成一條沉淀線，不同的抗原抗體系統(tǒng)在瓊脂中擴(kuò)散的速度不同，可在瓊脂中形成不同的沉淀線。本法主要是用于檢測血清中各種免疫球蛋白、甲胎蛋白、乙肝表面抗原等。缺點(diǎn)是需要時(shí)間過長，靈敏度不高。
1、稱取一定量的瓊脂粉，按1%左右（0.8%～1.5%）的比例加入生理鹽水或緩沖液，水浴煮沸融化20min。
2、將融化的瓊脂倒入平皿內(nèi)，使厚度2mm～3mm。自然冷卻。
3、根據(jù)要求（孔徑即孔的直徑，孔距即兩孔圓心之間的距離，包括兩孔的半徑）按模板打孔。也可直接用組合打孔器打孔?，F(xiàn)在一般多打成梅花形孔圖。
4、挑出孔內(nèi)瓊脂，注意不要挑破孔緣。
5、在火焰上緩緩加熱，使孔底邊緣的瓊脂少許熔化，以封底，以免加樣后液體從孔底滲漏。
6、以毛細(xì)滴管吸取樣品加入孔內(nèi)，注意不要產(chǎn)生氣泡，以加滿為度。加少了，影響反應(yīng)程度，加多了，易溢出，也影響反應(yīng)結(jié)果。
